RNA in situ hybridization assay specificity

Human PC9 lung carcinoma cells were used to determine the ViewRNA Assay specificity. PC9 cells were labeled with CellTracker® Green and were counted before and after the assay. In addition, PC9 cells were probed for KRT19 mRNA, and the number of KRT19 positive cells was compared with the number of CellTracker Green positive cells. The assay was highly specific in detecting green labeled PC9 cells (representing CTCs) expressing the KRT19 biomarker with an average specificity of 97% (Figure 1, Table 2). Read More ...


Detection of HER2 in human breast carcinoma SK-BR-3 cells

HER2 gene is known to be amplified in human breast SK-BR-3 cells. SK-BR-3 cells were spiked in 3 mL human whole blood and captured by the ScreenCell filter, followed by ViewRNA Assay using probes specific to HER2 (green), KRT19 (red), and CD45 (magenta). The results showed a high number of dots (puncta) per cell for HER2 and KRT19, demonstrating high RNA expression for both genes in SK-BR-3 cells.
